Using Isocratic Preparative Liquid Chromatography as a Quick First-Step Purification for Natural Product Isolation

چکیده
IN T RO DU C T IO N Preparative chromatography is the technique of choice for isolation and purification of different types of compounds, including pharmaceutical drug candidates, target compounds from synthetic mixtures, and bioactive compounds from natural products. As a result, there has been continuous research interest in improving the fundamental knowledge and instrumentation of preparative chromatography, with an ultimate goal of obtaining pure target compounds efficiently and cost-effectively.

Improving Resolution and Column Loading Systematically in Preparative Liquid Chromatography for Isolating a Minor Component from Peppermint Extract
■ At-column dilution alleviates the peak distortion and loss of resolution attributed to the injection of large volumes of strong solvent, leading to improved resolution, column loading, and overall productivity in natural product isolation. IN T RO DU C T IO N Natural products are widely used in the pharmaceutical, food supplement, nutraceutical, and alternative medicine industries. Chromatogr...

A preparative high-speed counter-current chromatography (HSCCC) method for isolation and purification of flavonoids from the Chinese medicinal plant Epimedium koreamum Nakai was successfully established by using chloroform-methanol-water (4:3.5:2, v/v) as the two-phase solvent system. The method yielded 11.4 mg of epimedokoreanoside I, 46.5 mg of icariin and 17.7 mg of icariside II from 200 mg ...

There is no other naturally occurring defense agent against cancer that has a stronger effectthan paclitaxel, commonly known under the brand name of Taxol®. The major drawback for themore widespread use of paclitaxel and its precious precursor, 10-deacetylbaccatin III (10-DABIII), is that they require large-scale extraction from different parts of yew trees (Taxus species),cell cultures, taxane...
متن کاملApplication of high-speed counter-current chromatography coupled with high-performance liquid chromatography-diode array detection for the preparative isolation and purification of hyperoside from Hypericum perforatum with online purity monitoring.
Following preparative isolation and purification by high-speed counter-current chromatography (HSCCC), the collected fractions were generally analyzed by high-performance liquid chromatography (HPLC) to determine the relative purities of each fraction. Our paper reports for the first time a preparative isolation-purity detection hyphenated system: online coupling of HSCCC with high-performance ...
